About PNC-27
p53 N-terminal / HDM2-targeting peptide
Research background as published in the scientific literature. Supplied for in-vitro laboratory research only; nothing here is a claim about use in humans or animals.
PNC-27 is a synthetic peptide comprising the MDM2-binding domain of the p53 tumour suppressor protein fused to a membrane-targeting leader sequence. It was developed as a research tool to investigate selective cancer cell targeting via HDM2 (human MDM2 homolog) which is overexpressed on the surface of many cancer cell types.
Preclinical studies have reported that PNC-27 induces membrane disruption selectively in cancer cell lines while leaving normal cells unaffected. This selectivity profile makes it a valuable compound for investigating cancer membrane biology and targeted apoptosis mechanisms.

- 01HDM2 / MDM2 InteractionPNC-27 is derived from the MDM2-binding domain of p53. Research has examined how it interacts with HDM2 proteins overexpressed on cancer cell membranes, and whether this interaction can selectively target malignant cells while sparing normal cells.
- 02Membrane Disruption SelectivityLaboratory studies have investigated PNC-27's selective disruption of cancer cell membranes. The mechanism is proposed to involve binding to membrane-expressed HDM2, leading to pore formation and cell death selectively in HDM2-overexpressing cells.
- 03p53 Tumour Suppressor PathwayResearch has explored PNC-27's interaction with the p53-MDM2 signalling axis. Studies examine how modulation of this pathway influences apoptotic signalling in cancer cell line models versus normal cell controls.
