About FOX04-DRI
FOXO4-D-Retro-Inverso Peptide
Research background as published in the scientific literature. Supplied for in-vitro laboratory research only; nothing here is a claim about use in humans or animals.
FOX04-DRI (FOXO4-D-Retro-Inverso) is a peptidomimetic designed to interfere with the interaction between the FOXO4 transcription factor and p53 in senescent cells. Senescent cells accumulate with age and contribute to tissue dysfunction through the secretion of pro-inflammatory factors (SASP).
Research published in Cell (2017) by Baar et al. demonstrated that FOX04-DRI could selectively induce apoptosis in senescent cells while sparing healthy cells, with treated aged mice showing improvements in physical function, renal function, and fur density. This represented a significant advance in the senolytic peptide research field.

- 01Senescent Cell ClearanceFOX04-DRI is designed to disrupt the interaction between FOXO4 and p53 in senescent cells, enabling p53-mediated apoptosis. Research examines its ability to selectively clear senescent cells without affecting healthy proliferating cells.
- 02p53 Apoptotic PathwayResearch has investigated how disrupting the FOXO4-p53 interaction releases p53 to translocate to mitochondria and initiate intrinsic apoptosis specifically in senescent cells that rely on this interaction for survival.
- 03Physiological RejuvenationPreclinical studies have examined whether clearance of senescent cell burden using FOX04-DRI restores tissue function, physical performance, and biomarker profiles associated with ageing in murine models.
