About PE-22-28
PE-22-28 (Spadin analogue)
Research background as published in the scientific literature. Supplied for in-vitro laboratory research only; nothing here is a claim about use in humans or animals.
PE-22-28 is a synthetic peptide derived from the propeptide of NTSR3/sortilin and is an optimised analogue of Spadin, a natural TREK-1 antagonist. TREK-1 is a two-pore domain (K2P) potassium channel expressed in neurons that regulates resting membrane potential.
Research interest in TREK-1 antagonists increased when TREK-1 knockout mice showed antidepressant-like phenotypes. PE-22-28 was developed as an improved Spadin analogue with enhanced stability and potency, and has been studied in preclinical antidepressant models for its effects on serotonin neurotransmission and behavioural outcomes.

- 01TREK-1 Potassium Channel AntagonismPE-22-28 is a synthetic optimised analogue of Spadin that acts as an antagonist of the TREK-1 (KCNK2) two-pore domain potassium channel. Research has examined how TREK-1 blockade modulates neuronal excitability and serotonergic transmission.
- 02Serotonin System ModulationResearch has investigated PE-22-28's effects on serotonergic neurotransmission, with studies examining whether TREK-1 antagonism increases serotonin availability in relevant brain regions through enhanced neuronal firing rates.
- 03Rapid-Onset ResearchA key research interest in TREK-1 antagonists is their potential for rapid-onset antidepressant-like effects in preclinical models. Research has examined PE-22-28 in forced swim test and tail suspension models, with studies reporting faster onset than monoamine-based comparators.
