About Melanotan 1

Melanotan I (afamelanotide)

Research background as published in the scientific literature. Supplied for in-vitro laboratory research only; nothing here is a claim about use in humans or animals.

Melanotan I (also known as afamelanotide) is a synthetic linear analogue of alpha-melanocyte-stimulating hormone (alpha-MSH) developed at the University of Arizona. Unlike Melanotan II, it is a linear peptide with higher selectivity for MC1R over other melanocortin receptor subtypes.

Afamelanotide (the clinical form of Melanotan I) received EMA approval in 2014 and TGA approval in Australia for the treatment of erythropoietic protoporphyria (EPP), making it one of the few melanocortin peptides to reach the market. Research continues to examine its photoprotective properties and applications in other photodermatological conditions.

  1. 01MC1R Selective AgonismMelanotan I is a synthetic linear analogue of alpha-MSH with high selectivity for MC1R. Research has examined its role in stimulating melanocyte differentiation, melanin synthesis, and eumelanin upregulation in skin models.
  2. 02Photoprotection ResearchResearch has examined whether increased melanin production induced by Melanotan I reduces UV-induced DNA damage in skin cell models. This formed the basis for clinical development of afamelanotide as a photoprotective agent.
  3. 03Erythropoietic ProtoporphyriaMelanotan I (as afamelanotide) has progressed furthest clinically, with regulatory approval in Europe and Australia for erythropoietic protoporphyria (EPP). Research has examined its effects on phototoxic reactions and quality of life in EPP patients.
